AD is characterized by a pathognomonic early decrease in brain blood flow, the
accumulation of the Abeta peptide in the wall of brain vessels (amyloid angiopathy), and
a high local inflammation surrounding senile plaques. These three circumstances lead to a
local decrease in oxygen concentration that could modify the progression of AD. Indeed,
we have shown that human AD brains show molecular indications of hypoxia. We have
also revealed that the accumulation of HIFalpha factors produces microglia cell cycle
arrest and worsens AD pathology in animal models of AD. We have identified PHD3, a HIF
target gene, as another contributor to AD neurodegeneration. Genetic deletion of PHD3
recovers cognitive and behavioral phenotypes of an AD mouse model and change the
activation profile of microglia.

Graduated in Biology in 1994 with maximal academic honors, the PI of the present project
worked in his PhD for 4 years (1995-1998), with a fellowship from the Spanish Ministry for
Science and Education. He published 4 papers as a first author with only his PhD
supervisor as a second author (corresponding) in journals of medium-high impact like
PNAS. The PI moved to “Institut Alfred Fessard” at Gif-sur-Yvette, France where
performed a postdoctoral stage of 4 years (1999-2002, EMBO and FRM fellowships),
under the supervision of Dr. Thomas Préat. He published five papers as a first author

including
two Science and one Nature article. The PI obtained a competitive contract to

return to Spain (Ramón y Cajal, Spanish Ministry of Science) associated with Prof. LópezBarneo in a Hospital in Seville (2003). During this time, the PI got a permanent position at
the Spanish Research Council (CSIC) to become an independent researcher (2007). He was
selected as PI of the Instituto de Biomedicina de Sevilla (IBiS) during 2008 and gained
research space in 2011. His main line of research is related with Alzheimer’s disease (AD)
and the hypoxia-signaling pathway.
